What is it that shapes our microbiome? A study brings new hope for improving our health — ScienceDaily

 

The question of nature vs nurture extends to our microbiome — the personal complement of mostly-friendly bacteria we carry around with us. Study after study has found that our microbiome affects nearly every aspect of our health; and its microbial composition, which varies from individual to individual, may hold the key to everything from weight gain to moods. Some microbiome researchers had suggested that this variation begins with differences in our genes; but a large-scale study conducted at the Weizmann Institute of Science challenges this idea and provides evidence that the connection between microbiome and health may be even more important than we thought.

Indeed, the working hypothesis has been that genetics plays a major role in determining microbiome variation among people. According to this view, our genes determine the environment our microbiome occupies, and each particular environment allows certain bacterial strains to thrive. However, the Weizmann researchers were surprised to discover that the host’s genetics play a very minor role in determining microbiome composition — only accounting for about 2% of the variation between populations.

The research was led by research students Daphna Rothschild, Dr. Omer Weissbrod and Dr. Elad Barkan from the lab of Prof. Eran Segal of the Computer Science and Applied Mathematics Department, together with members of Prof. Eran Elinav’s group of the Immunology Department, all at the Weizmann Institute of Science. Their findings, which were recently published in Nature, were based on a unique database of around 1,000 Israelis who had participated in a longitudinal study of personalized nutrition. Israel has a highly diverse population, which presents an ideal experimental setting for investigating the effects of genetic differences. In addition to genetic data and microbiome composition, the information collected for each study participant included dietary habits, lifestyle, medications and additional measurements. The scientists analyzing this data concluded that diet and lifestyle are by far the most dominant factors shaping our microbiome composition.

If microbiome populations are not shaped by our genetics, how do they nonetheless interact with our genes to modify our health? The scientists investigated the connections between microbiome and the measurements in the database of cholesterol, weight, blood glucose levels, and other clinical parameters. The study results were very surprising: For most of these clinical measures, the association with bacterial genomes was at least as strong, and in some cases stronger, than the association with the host’s human genome.

According to the scientists, these findings provide solid evidence that understanding the factors that shape our microbiome may be key to understanding and treating many common health problems.

Dietary fibers promote gut bacteria that benefit blood glucose control

 

The fight against type 2 diabetes may soon improve thanks to a pioneering high-fiber diet study led by a Rutgers University-New Brunswick professor.

Promotion of a select group of gut bacteria by a diet high in diverse fibers led to better blood glucose control, greater weight loss and better lipid levels in people with type 2 diabetes, according to research published today in Science.

The study, underway for six years, provides evidence that eating more of the right dietary fibers may rebalance the gut microbiota, or the ecosystem of bacteria in the gastrointestinal tract that help digest food and are important for overall human health.

“Our study lays the foundation and opens the possibility that fibers targeting this group of gut bacteria could eventually become a major part of your diet and your treatment,” said Liping Zhao, the study’s lead author and a professor in the Department of Biochemistry and Microbiology, School of Environmental and Biological Sciences at Rutgers University-New Brunswick.

Type 2 diabetes, one of the most common debilitating diseases, develops when the pancreas makes too little insulin — a hormone that helps glucose enter cells for use as energy — or the body doesn’t use insulin well.

In the gut, many bacteria break down carbohydrates, such as dietary fibers, and produce short-chain fatty acids that nourish our gut lining cells, reduce inflammation and help control appetite. A shortage of short-chain fatty acids has been associated with type 2 diabetes and other diseases. Many clinical studies also show that increasing dietary fiber intake could alleviate type 2 diabetes, but the effectiveness can vary due to the lack of understanding of the mechanisms, according to Zhao, who works in New Jersey Institute for Food, Nutrition, and Health at Rutgers-New Brunswick.

In research based in China, Zhao and scientists from Shanghai Jiao Tong University and Yan Lam, a research assistant professor in Zhao’s lab at Rutgers, randomized patients with type 2 diabetes into two groups. The control group received standard patient education and dietary recommendations. The treatment group was given a large amount of many types of dietary fibers while ingesting a similar diet for energy and major nutrients. Both groups took the drug acarbose to help control blood glucose.

The high-fiber diet included whole grains, traditional Chinese medicinal foods rich in dietary fibers and prebiotics, which promote growth of short-chain fatty acid-producing gut bacteria. After 12 weeks, patients on the high-fiber diet had greater reduction in a three-month average of blood glucose levels. Their fasting blood glucose levels also dropped faster and they lost more weight.

Surprisingly, of the 141 strains of short-chain fatty acid-producing gut bacteria identified by next-generation sequencing, only 15 are promoted by consuming more fibers and thus are likely to be the key drivers of better health. Bolstered by the high-fiber diet, they became the dominant strains in the gut after they boosted levels of the short-chain fatty acids butyrate and acetate. These acids created a mildly acidic gut environment that reduced populations of detrimental bacteria and led to increased insulin production and better blood glucose control.

The study supports establishing a healthy gut microbiota as a new nutritional approach for preventing and managing type 2 diabetes.

Modulation of Fgf21 gene in early-life ameliorates adulthood diet-induced obesity

 

The importance of good nutrition in the early development of children has been recognized for many decades. Nutritional experiences in early life can have profound and long-lasting effects on body weight in later life. For instance, malnutrition in early life as a result of poor nutrition during pregnancy and/or the lactation period may be stored on the offspring genome as epigenetic memory and persist into adulthood, thereby increasing the susceptibility to metabolic diseases such as obesity in later life. This area of epigenetics has become one of the fastest-growing and most complex areas of biological science.

A team of researchers led by Tokyo Medical and Dental University (TMDU) previously found that upon the onset of lactation after birth, milk lipids serve as a ligand to activate the nuclear receptor peroxisome proliferator-activated receptor (PPAR)α, which is a key transcriptional regulator of liver fat metabolism. They also demonstrated that administration of a synthetic PPARα ligand to mouse dams during the perinatal period induces enhanced reductions in DNA methylation of fatty-acid β-oxidation genes in the liver of the offspring. DNA methylation is an epigenetic mechanism used by cells to control gene expression.

These findings prompted the researchers to explore whether DNA methylation status of PPARα target genes, which is modulated and established in a PPARα-dependent manner in early life, persists into adulthood.

“In the current study, using a genome-wide analysis of DNA methylation, we identified a few PPARα target genes that underwent ligand-activated PPARα-dependent DNA demethylation during the perinatal period and whose DNA methylation status persists into adulthood,” explains a corresponding author Koshi Hashimoto . “We provide the first evidence that the PPARα-dependent gene Fgf21 demethylation occurs in the postnatal mouse liver, and once established it persists into adulthood and exerts long-term effects on the magnitude of gene expression response to environmental cues, which may account in part for the attenuation of diet-induced obesity (Figure).”

This study represents the first detailed analysis of DNA methylation status of a particular gene throughout life, and the results were recently published in Nature Communications.

“Given that PPARα may act as a sensor of milk lipids during the suckling period, it is likely that the suckling period provides a critical time window for PPARα-dependent Fgf21 demethylation in response to the maternal environment,” adds senior author Yoshihiro Ogawa. “We propose that Fgf21 methylation represents a form of epigenetic memory that persists into adulthood, and it may have a role in the developmental programming of obesity.”

Fish the primary source of nutrition in medieval Northern Ostrobothnia

 

Researchers investigated the diet of people buried in the Ii Hamina cemetery from the 15th to the 17th centuries by analysing isotopes in the bones of the deceased. Isotopes preserve information on the various nutrient sources used by humans during their lifetime. A study published in the Environmental Archaeology journal reveals that the dominant protein source was small fish, such as roach or Baltic herring.

The medieval cemetery of Ii Hamina is located next to the centre of the Ii municipality. Through investigations conducted at the cemetery, significant knowledge has been gained on past human generations in Northern Ostrobothnia and in Finland in general.

The study of the diet of medieval Ii residents indicated a very large share of fish-based food. Of all protein consumed, as much as 70% may have been fish. On the one hand, this is evidence of the importance of waterways; on the other hand, it indirectly indicates the insignificance of farming and dairying in the region.

Sufficient but unbalanced nutrition

A previous study already revealed that medieval residents of Ii had no significant trouble finding food.

“This new study confirms the notion that the diet in Ii was very likely sufficient,” says researcher Maria Lahtinen from the Finnish Museum of Natural History Luomus, part of the University of Helsinki.

The recently published study indicates that the fish consumed by the residents of Ii was probably from the middle of the food web, in other words roach, Baltic herring or other species feeding on benthic and other invertebrates. Species-specific findings cannot, however, be gained through isotope analysis, so the species mentioned are based on guesswork.

Seal hunting, on the other hand, most likely did not play a significant role in medieval Ii, the study finds.

In prior studies, the dental health of the deceased has also been investigated, revealing a very protein-rich diet compared to today. On average, the population at that time was also shorter. These factors are evidence of an unbalanced diet.

Another finding in the new study was an individual whose bone isotope consistency differed from others. The diet of this individual was much closer to living habits based primarily on farmed food. The bones of altogether 98 buried individuals were analysed in the study, which makes it very likely that this individual was originally from somewhere else or in some way enjoyed a special status in the community.

New diabetes drug may help people with obesity lose weight

 

A compound that mimics a naturally occurring hormone that regulates appetite may help people who have obesity but not diabetes to lose weight, a new study suggests. The research will be presented Sunday, March 18, at ENDO 2018, the Endocrine Society’s 100th annual meeting in Chicago, Ill.

The compound, semaglutide, has a chemical structure that is very similar to the hormone glucagon-like peptide 1 (GLP-1), which regulates both insulin secretion and appetite. In December, the U.S. Food and Drug Administration approved the semaglutide injection Ozempic as a once-weekly adjunct to diet and exercise to improve glycemic control in adults with type 2 diabetes.

“This randomized study of weight loss induced with semaglutide in people with obesity but without diabetes has shown the highest weight reductions yet seen for any pharmaceutical intervention,” said lead author Patrick M. O’Neil, Ph.D., Director of the Weight Management Center and Professor of Psychiatry and Behavioral Sciences at the Medical University of South Carolina in Charleston, S.C.

The new study included 957 participants, 35 percent of whom were male. All participants had a body mass index (BMI) of at least 30, but did not have diabetes. They were randomly assigned to seven different groups. Five groups received different doses of semaglutide (between 0.05 mg and 0.4 mg) via injection once daily; a sixth group received a placebo, and a seventh group received 3 mg of the diabetes drug liraglutide. All participants received monthly diet and exercise counseling.

After one year, all participants receiving semaglutide had lost significantly more weight than those receiving placebo. The higher the dose participants received, the greater their average weight loss. Participants who received 0.05 mg of semaglutide daily lost an average of 6.0 percent of their body weight; the 0.1 mg group lost an average of 8.6 percent; the 0.3 mg group lost an average of 11.2 percent; and those receiving a daily dose of 0.4 mg lost an average of 13.8 percent. Those receiving liraglutide lost an average of 7.8 percent of their body weight, while those in the placebo group lost only 2.3 percent on average.

Sixty five percent of participants who received 0.4 mg of semaglutide per day lost at least 10 percent of their body weight, compared with 10 percent of those in the placebo group and 34 percent of the liraglutide group.

The most common adverse events in those taking semaglutide were mild/moderate nausea, as seen previously with GLP-1 receptor agonists.

O’Neil noted that further studies of semaglutide for obesity are underway.
